Methods and Assays for Subtyping Staphylococcus Aureus Clonal Complex Eight Strains
Description
Distinguishing among the sub-lineages of Staphylococcus aureus clonal complex 8 (CC8) is critical for purposes of epidemiology and surveillance, especially as the epidemiologic separation between hospital-acquired (HA) and community-associated (CA) strains disappear. The technology is a new typing scheme for CC8 isolates that is rapid and easy to use. Also, the technology is based on stable genomic markers. The assay uses real-time PCR (polymerase chain reaction) and a labeled probe for detection.

Benefits and advantages
The assay allows for the identification of canonical single nucleotide polymorphisms (canSNPs) from a Staphylococcus aureus CC8 phylogeny to target each of the major lineages. The present invention involves a canSNP-based approach to eliminate the lineage confusion observed with pulse field gel electrophoresis (PFGE), spa typing, and mobile genetic marker typing, as SNPs are inherently stable and quantify relatedness among strains. The assay employs real-time PCR as a rapid, scalable technique that is common in public health and reference laboratories, making the assay an attractive tool for surveillance and epidemiology.
